Measuring immune response to 3D cultures

1.Establish 3D cell cultures in devices

2. Place in test chamber; open device

3.Pipet immune cells to test chamber

4.Assess immune cell migration and activity into devices by microscopy, flow cytometry, biochemical or histological approaches

PyVmT carcinoma cells were transfected with control or CCR2 siRNAs and cultured in TheraKan      devices, which were placed in test chambers containing mcherry+ Raw264.7 macrophages.  Devices were imaged for the presence of mcherry+ cells by fluorescence microscopy after 24 hours.
